I would definitely mention it to the director. I would say you are very much interested and explain why.
Did she definitely say she planned to look outside for the senior level associate? Or just that one was needed?

They are looking to add to the team now. They only do promotions at year end. So if they are planning on adding a senior associate they can only hire from outside. They will not promote internally at this time.
I feel like there's always a loophole.

I would address your interest in leading the team but I would NOT discuss any information that your co-worker overheard. Like LuckyStar mentioned did the person specifically say they were going to hire an external candidate? You do not want to appear unprofessional- so stick to the facts- the team is going to be expanding and you want to be the team leader- focus on showing your boss why they should give you the job.
