Bed and Breakfasts - Eastern LI?
Close to vineyards - any recs?
Preferrably none that are two-night minimums.

We stayed at the Mill House Inn in East Hampton a few years ago and it was fantastic, the breakfast was really yummy and so were the fresh cookies at night :)

We stayed here and it was very nice. I am not sure how far the wineries are.
